摘要

恶性黑色素瘤是最罕见但最具侵袭性和致命性的皮肤癌。黑色素瘤是黑色素细胞恶性转化的结果，导致其不受控制的增殖。丝裂原活化蛋白激酶(MAPK)通路的突变是其最常见的原因之一，它对细胞凋亡、分裂、生长、分化和迁移等细胞过程的控制至关重要。BRAF激酶作为该途径的已知靶点之一，多年来一直被认为是黑色素瘤治疗中的一个突出的分子靶点，下面的综述概述了有关其结构，突变和机制的最新知识。
