维胺酯通过调节S100A8/S100A9-MAPK级联抑制痤疮丙酸杆菌诱导的HaCat细胞异常增殖和角蛋白化

摘要

背景:维胺酯是我国开发的一种维生素a酸药物,已在临床上用于痤疮治疗,调节上皮细胞分化和增殖,抑制角质化,减少皮脂分泌,控制免疫和抗炎作用;然而,它的确切工作方法尚不清楚。 方法:采用痤疮丙酸杆菌联合皮脂涂抹法在大鼠耳内诱发痤疮。 结果:维胺酯治疗30天后,大鼠耳朵表皮增厚和角蛋白过度产生的症状明显改善。对大鼠皮肤组织的转录组学分析表明,维胺酯显著调节细胞角化的生物学途径。基因差异分析显示,在胺化处理后S100A8和S100A9基因显著下调。qPCR和Western印迹的结果证实,维胺酯在大鼠和HaCat细胞痤疮模型中抑制S100A8和S100A9基因和蛋白的表达,而其下游途径MAPK(MAPK p38/JNK/ERK1/2)蛋白表达水平受到抑制。额外施用S100A8和S100A9复合蛋白显著逆转了维胺酯对痤疮细胞模型中异常增殖和角质化水平的抑制作用。 结论:总之,维胺酯可以通过调节S100A8和S100A9来抑制MAPK通路的激活,抑制角质形成细胞的增殖和角质化水平,从而改善痤疮。
